Debido a su sofisticada tecnolog\u00eda gen\u00e9tica, as\u00ed como otras ventajas a nivel experimental, usamos los discos imaginales de la mosca de la fruta, Drosophila melanogaster, como sistema modelo para investigar los factores moleculares y gen\u00e9ticos que subyacen a la aparici\u00f3n de tumores. Nuestro trabajo se centra en tres l\u00edneas de investigaci\u00f3n.  <\/p>\n<p style=\"text-align: justify;\"><strong> <em> 1)El potencial tumorig\u00e9nico de c\u00e9lulas que no pueden ejecutar el programa apopt\u00f3tico<\/em>.<\/strong><\/p>\n<p style=\"text-align: justify;\">Esta l\u00ednea se basa en trabajos previos del laboratorio sobre el comportamiento de c\u00e9lulas en las cuales la maquinaria apopt\u00f3tica ha sido suprimida; estas c\u00e9lulas deficientes para la apoptosis (Apoptosis-Deficient, AD) sobreviven al estr\u00e9s o a los tratamientos con drogas que conducen a la muerte a las c\u00e9lulas normales. Hemos desarrollado un modelo de tumorog\u00e9nesis basado en las propiedades de estas c\u00e9lulas: tras inducir un estr\u00e9s transitorio una fracci\u00f3n de la poblaci\u00f3n de c\u00e9lulas AD activa ect\u00f3picamente las funciones pro-proliferativas y pro-migratorias de la v\u00eda de la Jun N-Terminal Kinase (JNK), lo cual resulta en la generaci\u00f3n de tumores invasivos. La generaci\u00f3n de estos tumores est\u00e1 mediada por la actividad de la JNK. Hemos visto que presentan caracter\u00edsticas de c\u00e9lulas senescentes, incluyendo el fenotipo secretor asociado a la senescencia (SASP). En particular, \u00e9stas c\u00e9lulas secretan mol\u00e9culas se\u00f1alizadoras como Dpp, Wg y Upd encargadas de activar las rutas de Dpp, Wg y JAK\/STAT respectivamente. Experimentos en los cuales se han suprimido estas v\u00edas indican que su funci\u00f3n es responsable de los sobrecrecimientos tumorales, aunque se desconocen los efectos espec\u00edficos de cada una de las v\u00edas as\u00ed como las interacciones entre ellas que est\u00e1n siendo objeto de estudio en el laboratorio. Tenemos evidencias preliminares de que no todas las c\u00e9lulas senescentes adquieren el fenotipo SASP; aquellas con SASP aparecen en la regi\u00f3n central del disco, mientras que las localizadas en el notum o en regiones perif\u00e9ricas no presentan SASP. La observaci\u00f3n de que existen c\u00e9lulas senescentes SASP y no-SASP es nueva en el campo. Correlaciona con la observaci\u00f3n de que los crecimientos tumorales ocurren en la regi\u00f3n central del disco, lugar d\u00f3nde se concentran las c\u00e9lulas senescentes secretoras. El mecanismo por el cual se produce este efecto dependiente de posici\u00f3n est\u00e1 siendo investigado en el laboratorio.       <\/p>\n<p style=\"text-align: justify;\">Un aspecto relacionado es c\u00f3mo la actividad de la JNK es capaz de ejercer todas estas funciones. Hace unos a\u00f1os llevamos a cabo una b\u00fasqueda por RNAseq de genes espec\u00edficamente activados por la v\u00eda de la JNK y necesarios para generar sobrecrecimientos. Algunos de los genes identificados est\u00e1n siendo investigados por su implicaci\u00f3n con las se\u00f1ales que emanan de las c\u00e9lulas senescentes y los sobrecimientos asociados.  <\/p>\n<p style=\"text-align: justify;\"><strong><em>2) Modificaciones de la cromatina en c\u00e9lulas senescentes y reversibilidad de la senescencia<\/em><\/strong><\/p>\n<p style=\"text-align: justify;\">La senescencia puede ser inducida por uno o varios est\u00edmulos que producen estr\u00e9s celular (como, por ejemplo, da\u00f1o al ADN, estr\u00e9s oxidativo o activaci\u00f3n de oncogenes). Todos ellos producen alteraciones, generalmente estables, en la funci\u00f3n y estructura de la c\u00e9lula. Uno de los marcadores de la transformaci\u00f3n es un cambio en la estructura de la cromatina, denominado \u201csenescence-associated heterochromatin foci (SAHFs)\u201d es decir, focos de heterocromatina asociada a senescencia. En c\u00e9lulas de mam\u00edferos, tras la inducci\u00f3n de senescencia aparecen los SAHFs como focos enriquecidos en marcadores de heterocromatina (como H3K9me3) y con niveles disminuidos de marcadores eucrom\u00e1ticos. En el laboratorio estamos empezando a describir el proceso por el cual las c\u00e9lulas entran en senescencia, algo que no ha sido descrito hasta la fecha en Drosophila. Hemos visto que, an\u00e1logamente a lo que ocurre en c\u00e9lulas de vertebrados en cultivo, las c\u00e9lulas del disco de ala de Drosophila aumentan el tama\u00f1o de sus nucl\u00e9olos al volverse senescentes. Los niveles de H3K9me3 tambi\u00e9n aumentan en esas c\u00e9lulas. Dado que la v\u00eda de la JNK es un inductor de senescencia, estamos estudiando c\u00f3mo act\u00faa sobre las modificaciones en cromatina. Algunas de las dianas de JNK identificadas en la b\u00fasqueda de ARNs mencionada anteriormente est\u00e1n asociadas con modificaciones cromat\u00ednicas (como Caf1-105, involucrado en la formaci\u00f3n y el mantenimiento de la heterocromatina).        Estamos analizando las posibles implicaciones de algunos de esos genes en la remodelaci\u00f3n de la cromatina asociada con la senescencia.<\/p>\n<p style=\"text-align: justify;\">Adem\u00e1s, hemos encontrado un funci\u00f3n inesperada de los transposones F-element (pertenecientes a la familia LINE) en el mantenimiento del estado de senescencia; al expresarlos a altos niveles las c\u00e9lulas senescentes parecen revertir a su condici\u00f3n silvestre (figura 2). Se trata de una observaci\u00f3n de gran inter\u00e9s, en primer lugar porque sugiere que estos transposones-que ya se ha visto que tienen funciones en la remodelaci\u00f3n de la cromatina durante los primeros estadios embrionarios tanto en vertebrados como en <span> <\/span><em>Drosophila<\/em><span> <\/span>-funcionan tambi\u00e9n en senescencia, y en segundo lugar porque es el primer indicio descrito de que la senescencia puede ser revertida. Los detalles moleculares de este fen\u00f3meno est\u00e1n actualmente siendo investigados. <\/p>\n<p style=\"text-align: justify;\"><strong><em>3) Interacciones entre poblaciones celulares con distintas tasas de proliferaci\u00f3n<\/em><\/strong><\/p>\n<p style=\"text-align: justify;\">En nuestros an\u00e1lisis de tumores producidos por mutaciones oncog\u00e9nicas en<span> <\/span><em>polyhomeotic<\/em><span> <\/span>(<em>(ph)<\/em>) y<span> <\/span><em>erupted<\/em><span> <\/span>(el<span> <\/span><em>Drosophila<\/em><span> <\/span>hom\u00f3logo en Drosophila del gen<span> <\/span><em>tumor susceptibility gene<\/em><span> <\/span><em>101<\/em>), hemos realizado observaciones que pudieran tener implicaciones a la hora de entender c\u00f3mo las c\u00e9lulas tumorales colonizan tejidos adyacentes: en experimentos en los cuales un compartimento del disco de ala es silvestre y el otro sobrecrece debido a que es mutante para el oncogen<span> <\/span><em>ph<\/em><span> <\/span>o<span> <\/span><em>erupted<\/em><span> <\/span>el compartimento silvestre crece menos que su tama\u00f1o en condiciones normales<\/p>\n<p style=\"text-align: justify;\">Hemos observado que este fen\u00f3meno ocurre tambi\u00e9n entre regiones del mismo compartimento. Lo que parece estar ocurriendo es que la poblaci\u00f3n de c\u00e9lulas que sobrecrece inhibe el crecimiento de los tejidos vecinos. Nos referimos a esas poblaciones como \u201cganadora\u201d (la que sobrecrece) y \u201cperdedora\u201d (la que pierde tama\u00f1o). Hemos descartado la hip\u00f3tesis de que la poblaci\u00f3n perdedora sufra un incremento de muerte por apoptosis, pero hemos visto que esta poblaci\u00f3n tiene un \u00edndice mit\u00f3tico menor de lo normal, lo cual explica su crecimiento reducido. Dado que el crecimiento general del disco depende de factores de crecimiento se\u00f1alizadores como Dpp o Wg, estamos estudiando la posibilidad de que las c\u00e9lulas de la poblaci\u00f3n ganadora secuestren las mol\u00e9culas se\u00f1alizadoras Dpp o Wg impidiendo su accesiblidad a la poblaci\u00f3n perdedora.    <\/p>\n<p style=\"text-align: justify;\">Tambi\u00e9n estamos tratando de identificar los factores gen\u00e9ticos asociados con la reducci\u00f3n del \u00edndice mit\u00f3tico, examinando los niveles de expresi\u00f3n de genes reguladores de la divisi\u00f3n celular, tanto los que la promueven (<em>cdk<\/em>, y otras quinasas dependientes de ciclinas) como las que la inhiben (<em>dacapo, frs<\/em>). Existe tambi\u00e9n la posibilidad de que la via de Hippo, un inhibidor de la proliferaci\u00f3n celular, est\u00e9 alterada, lo cual podr\u00eda testarse usando numerosos marcadores disponibles para
