El laboratorio de la Dra. Revilla estudia distintos aspectos moleculares de la interacci\u00f3n del virus de la peste porcina africana (VPPA) con su c\u00e9lula diana, abordando los mecanismos del tropismo, los genes de virulencia y el control viral del sistema inmune. Esto nos ha permitido la generaci\u00f3n de candidatos vacunales mediante CRISPR\/Cas9. El VPPA representa la mayor pandemia animal en todo el mundo, y urge el desarrollo de vacunas seguras y eficaces. <\/p>\n

El laboratorio del Virus de la peste porcina africana (VPPA) dirigido por la Dr. Yolanda Revilla, estudia los complejos mecanismos virales y celulares que se producen durante la infecci\u00f3n del VPPA. La c\u00e9lula diana natural de este virus son exclusivamente los monocitos y macr\u00f3fagos porcinos, por lo que los factores tanto virales como celulares que determinan su tropismo son un foco de atenci\u00f3n en nuestro laboratorio. Otro de los objetivos relevante lo constituyen los mecanismos moleculares implicados en la virulencia del VPPA, que est\u00e1n directamente relacionados con el control que el virus ejerce sobre distintas ramas de la respuesta inmune del animal infectado, y, en particular, sobre la respuesta innata regulando el IFN de tipo I, como hemos descrito en el grupo. Adem\u00e1s, estamos interesados en generar candidatos vacunales frente al VPPA basados, fundamentalmente, en vacunas vivas atenuadas (LAVs) y muy recientemente, tambi\u00e9n basados en prototipos s\u00f3lo parcialmente replicativos, lo que constituye una primicia en el campo y ser\u00eda un gran avance por su seguridad. Para ello, abordamos el estudio de la interacci\u00f3n virus-hu\u00e9sped mediante t\u00e9cnicas de biolog\u00eda celular, bioqu\u00edmicas, microscop\u00eda y citometr\u00eda, junto con la generaci\u00f3n de virus recombinantes mediante la tecnolog\u00eda CRISPR-Cas9 (adaptada a la biolog\u00eda del virus por el Dr. Daniel P\u00e9rez N\u00fa\u00f1ez, postdoc senior en el grupo), as\u00ed como su caracterizaci\u00f3n gen\u00e9tica con t\u00e9cnicas NGS como Illumina y Nanopore. Colaboramos adem\u00e1s activamente con otros centros de investigaci\u00f3n y empresas, tanto en Espa\u00f1a como en Europa y Asia, para analizar y evaluar el comportamiento del VPPA in vivo, as\u00ed como la respuesta inmune protectora de los distintos prototipos vacunales. <\/p>\n

El VPPA infecta cerdos dom\u00e9sticos y jabal\u00edes y constituye en la actualidad la gran pandemia animal, siendo el mayor problema de la industria porcina mundial adem\u00e1s de ser el responsable de un gran desastre ecol\u00f3gico. Desde 2007 un brote en el C\u00e1ucaso, el VPPA se fue extendiendo por Rusia hacia Europa del este, y en la actualidad est\u00e1 muy extendido por Europa, Asia (en particular China) y \u00c1frica, donde es end\u00e9mico, adem\u00e1s de estar presente en Ocean\u00eda y el Caribe, amenazando seriamente con continuar su expansi\u00f3n por Am\u00e9rica. Los estudios moleculares de la interacci\u00f3n virus-c\u00e9lula, as\u00ed como la generaci\u00f3n de prototipos vacunales, contribuir\u00e1n en el medio y largo plazo a contener y prevenir la gran amenaza que en la actualidad supone el VPPA. <\/p>\n
