El acortamiento de los tel\u00f3meros durante la divisi\u00f3n celular puede causar fusiones telom\u00e9ricas e inestabilidad gen\u00f3mica en tumores. Nuestro grupo investiga la formaci\u00f3n y funci\u00f3n de estas fusiones telom\u00e9ricas utilizando t\u00e9cnicas bioqu\u00edmicas, moleculares, CRISPR\/Cas y an\u00e1lisis gen\u00f3micos. El fin \u00faltimo de nuestra investigaci\u00f3n es el descubrimiento de nuevos marcadores y tratamientos contra el c\u00e1ncer <\/p>\n

Tel\u00f3meros en c\u00e1ncer y regeneraci\u00f3n<\/p>\n

Tel\u00f3meros en c\u00e1ncer y regeneraci\u00f3n. Los tel\u00f3meros son componentes esenciales de nuestro genoma ya que lo protegen de su deterioro y mantienen su estabilidad. Las fusiones telom\u00e9ricas ocurren cuando dos cromosomas se unen f\u00edsicamente a trav\u00e9s de sus extremos. Las fusiones telom\u00e9ricas pueden desencadenar una serie de reordenamientos gen\u00f3micos y alteraciones oncog\u00e9nicas que, en \u00faltima instancia, conducen al desarrollo de tumores malignos y a la resistencia a la quimioterapia. A pesar de su relevancia en la evoluci\u00f3n tumoral, nuestra comprensi\u00f3n de los patrones y consecuencias de las fusiones telom\u00e9ricas en el c\u00e1ncer humano sigue siendo limitada. Nuestro grupo est\u00e1 interesado en comprender la funci\u00f3n de las fusiones telom\u00e9ricas en c\u00e1ncer y regeneraci\u00f3n. <\/p>\n

Fusiones telom\u00e9ricas (FT) en c\u00e1ncer. En los \u00faltimos a\u00f1os hemos caracterizado las tasas y el espectro de las FT som\u00e1ticas en m\u00e1s de 30 tipos de c\u00e1ncer. Hemos descubierto que las FT est\u00e1n muy extendidas en los tumores humanos, con tasas que var\u00edan enormemente entre y dentro de los distintos tipos de c\u00e1ncer. Adem\u00e1s de FT de extremo a extremo, hemos identificado patrones \u00fanicos relacionados con la v\u00eda alternativa de alargamiento de los tel\u00f3meros (ALT). Un descubrimiento especialmente interesante ha sido la detecci\u00f3n de FT en la sangre de pacientes con c\u00e1ncer, lo que ofrece un m\u00e9todo potencial para la detecci\u00f3n precoz del c\u00e1ncer con alta especificidad y sensibilidad. Esto es especialmente prometedor para tipos de c\u00e1ncer como el de p\u00e1ncreas y los tumores cerebrales, que actualmente cuentan con opciones limitadas para su detecci\u00f3n precoz. Mediante t\u00e9cnicas bioqu\u00edmicas, moleculares, CRISPR\/Cas y gen\u00f3micas (NGS), seguiremos investigando la aparici\u00f3n y la funci\u00f3n de FT en diversos tumores humanos y de rat\u00f3n. Comprender el papel de las FT ser\u00e1 crucial para desarrollar terapias dirigidas en pacientes con c\u00e1ncer. <\/p>\n

Fusiones telom\u00e9ricas (TF) en la maduraci\u00f3n de los cardiomiocitos y en regeneraci\u00f3n cardiaca. La maduraci\u00f3n en muchas c\u00e9lulas de mam\u00edferos, incluidos los cardiomiocitos, va acompa\u00f1ada de un aumento del n\u00famero de cromosomas, o poliploidizaci\u00f3n. Dado que las c\u00e9lulas poliploides no logran dividirse en la mayor\u00eda de los casos, la aparici\u00f3n de la poliploid\u00eda puede explicar por qu\u00e9 los cardiomiocitos no proliferan lo suficiente para regenerar el coraz\u00f3n despu\u00e9s de un infarto. A pesar de su importancia, el mecanismo de poliploidizaci\u00f3n en cardiomiocitos sigue sin estar bien definido. En los \u00faltimos a\u00f1os, utilizando im\u00e1genes de alto contenido, hemos descubierto que aproximadamente la mitad de los cardiomiocitos postnatales de rat\u00f3n que entran en mitosis tienen puentes cromos\u00f3micos discernibles. Adem\u00e1s, observamos que la ploid\u00eda de los cardiomiocitos coincide con un n\u00famero creciente de FTs lo que sugiere una ruta directa a la poliploidizaci\u00f3n. FTs est\u00e1n presentes en los corazones de ratones, peces cebra, cerdos y humanos. Los corazones de pez cebra tienen relativamente pocas FTs, lo que correlaciona con que que sus cardiomiocitos son mayoritariamente diploides y su alta capacidad regenerativa. La eliminaci\u00f3n de la telomerasa inhibe la respuesta regenerativa y aumenta el n\u00famero de cardiomiocitos poliploides, lo que respalda la idea de que la poliploidizaci\u00f3n de los cardiomiocitos afecta a la regeneraci\u00f3n del coraz\u00f3n. En conjunto, estos resultados identifican a las FT como un mecanismo que conduce a la formaci\u00f3n de puentes cromos\u00f3micos y a la poliploidizaci\u00f3n, y sugieren que la citocinesis fallida de los cardiomiocitos se origina a partir de un defecto mit\u00f3tico. Actualmente estamos investigando si una regulaci\u00f3n precisa de la telomerasa evita la presencia de FTs, aumenta el n\u00famero de cardiomiocitos diploides y ampl\u00eda la ventana de regeneraci\u00f3n cardiaca. <\/p>\n
