Nuestro prop\u00f3sito es mejorar el control de las infecciones cr\u00f3nicas y oportunistas por el sistema inmunitario, usando nuevas plataformas tecnol\u00f3gicas que induzcan una buena respuesta de linfocitos T citot\u00f3xicos. En modelos de inmunodeficiencia parcial queremos, a largo plazo, mejorar el dise\u00f1o de nuevas vacunas, en especial para poblaciones vulnerables, como individuos inmunodeprimidos y ancianos. <\/p>\n

El objetivo del laboratorio se centra en mejorar el control del sistema inmunol\u00f3gico frente a infecciones cr\u00f3nicas y oportunistas. En estas situaciones, la respuesta inmune celular desempe\u00f1a un papel crucial, frente al papel de los anticuerpos en las infecciones agudas, que es la diana actual de las vacunas. El objetivo final es contribuir al dise\u00f1o racional de vacunas. A pesar del conocimiento acumulado, a\u00fan no disponemos de vacunas que generen una respuesta potente y duradera de los linfocitos T.
Nuestro trabajo tambi\u00e9n se enfoca en poblaciones vulnerables, como individuos inmunodeprimidos y ancianos. Buscamos mejorar la eficacia de las vacunas y medir la fuerza y durabilidad de la respuesta inmunitaria tras la vacunaci\u00f3n. Esto nos permite identificar par\u00e1metros cr\u00edticos de protecci\u00f3n mediada por la vacuna en estas poblaciones. <\/p>\n

Para ello, analizamos aspectos b\u00e1sicos de la respuesta inmunitaria celular antiviral y la vacunaci\u00f3n. Utilizamos modelos de rat\u00f3n con inmunodeficiencia parcial, combinados con la infecci\u00f3n por dos virus relevantes para el desarrollo de vacunas. Adem\u00e1s, exploramos nuevas plataformas tecnol\u00f3gicas que faciliten la respuesta celular mediante el transporte de ant\u00edgenos a los compartimentos celulares adecuados para su procesamiento.
Como modelo experimental, utilizamos ratones deficientes en el transportador asociado al procesamiento antig\u00e9nico (TAP). Este transportador desempe\u00f1a un papel central en la v\u00eda de presentaci\u00f3n de ant\u00edgenos a trav\u00e9s de mol\u00e9culas MHC de clase I (MHC-I), que son reconocidas por linfocitos T CD8+ citot\u00f3xicos espec\u00edficos de pat\u00f3genos. Estos linfocitos median en la eliminaci\u00f3n de las c\u00e9lulas infectadas in vivo.
El TAP se inactiva con frecuencia en las met\u00e1stasis y tambi\u00e9n es el objetivo de los virus. En ambos casos, esto da lugar a la evasi\u00f3n de la detecci\u00f3n y eliminaci\u00f3n por parte de los linfocitos T CD8+.
En nuestro laboratorio, estamos identificando ep\u00edtopos pept\u00eddicos virales que son presentados por el MHC-I en ausencia de TAP. Tambi\u00e9n investigamos las proteasas y las v\u00edas de procesamiento antig\u00e9nico que generan estos ep\u00edtopos en las c\u00e9lulas infectadas. Adem\u00e1s, evaluamos su contribuci\u00f3n a la respuesta CD8+ en estos animales.
Los modelos de infecci\u00f3n viral que utilizamos son el virus vaccinia y el citomegalovirus de rat\u00f3n. El virus vaccinia, un poxvirus como el mpox, es el vector vacunal del primer y mayor \u00e9xito en el uso de la vacunaci\u00f3n en salud p\u00fablica: la erradicaci\u00f3n de la viruela, declarada oficialmente en 1980. Los citomegalovirus son relevantes como potenciales vectores vacunales, ya que durante la infecci\u00f3n latente presentan ant\u00edgenos virales de forma continua, sin patolog\u00eda asociada, dando lugar a lo largo de la vida a un elevado n\u00famero de linfocitos T virus-espec\u00edficos, la llamada memoria inflacionaria.
Por \u00faltimo, en nuestro estudio continuado de la respuesta inmunitaria a los poxvirus, estamos explorando c\u00f3mo puede aplicarse esta informaci\u00f3n a nuevas enfermedades emergentes. Uno de los ejemplos m\u00e1s recientes es el brote de viruela del mono (Mpox). En particular, estamos estudiando la respuesta inmunitaria protectora en los ancianos que recibieron vacunas contra la viruela hace m\u00e1s de cincuenta a\u00f1os, y su capacidad para responder a nuevas amenazas v\u00edricas. <\/p>\n
