Javier Diez Guerra<\/strong> ha desarrollado CaMK2rep, un nuevo sensor biol\u00f3gico que permite <em>monitorizar<\/em> con gran sensibilidad la actividad de CaMKII. Esta prote\u00edna act\u00faa como un distribuidor \u00a0de se\u00f1ales dentro de las c\u00e9lulas nerviosas y est\u00e1 directamente relacionada con procesos tan importantes como la memoria, el aprendizaje y la salud del coraz\u00f3n.<\/p>\n<p>La prote\u00edna CaMKII es una enzima clave en la se\u00f1alizaci\u00f3n celular. Aunque se encuentra en todo tipo de tejidos, se expresa sobre todo en el cerebro y en el coraz\u00f3n. En el caso de las neuronas, act\u00faa como interruptor molecular, es decir, se activa cuando la concentraci\u00f3n de iones de calcio dentro de la c\u00e9lula aumenta r\u00e1pidamente como respuesta a un est\u00edmulo. Ello permite a las c\u00e9lulas responder a los cambios en su entorno, lo que es clave en procesos como la plasticidad neuronal. En este sentido, la actividad de CaMKII contribuye a fortalecer las conexiones neuronales al regular la morfolog\u00eda y el tama\u00f1o de las espinas dendr\u00edticas (las estructuras de las neuronas que reciben los impulsos nerviosos de otras neuronas).<\/p>\n<p>Sin embargo, hasta ahora, los cient\u00edficos y cient\u00edficas ten\u00edan pocas herramientas a su alcance para medir de manera precisa c\u00f3mo trabaja esta prote\u00edna en condiciones reales. Con el objetivo de superar esta limitaci\u00f3n, la nueva herramienta desarrollada en el CBM permite obtener m\u00e1s sensibilidad y una imagen m\u00e1s clara y fiable de su actividad.<\/p>\n<p>Esta nueva herramienta, denominada CaMK2rep y publicada en la revista <a href=\"https:\/\/pubs.acs.org\/doi\/10.1021\/acs.analchem.5c03227\"><em>Analytical Chemistry<\/em><\/a>, es un sensor biol\u00f3gico, es decir, un dispositivo producido por las propias c\u00e9lulas y fosforilable por CaMKII. El sensor utiliza los cambios en su fosforilaci\u00f3n como indicador de la actividad CaMKII.<\/p>\n<p>\u201cEste desarrollo responde a una necesidad real en la investigaci\u00f3n biom\u00e9dica: disponer de una herramienta sensible y fiable para cuantificar la actividad de CaMKII. Con ella podremos abordar preguntas clave en neurociencia y fisiopatolog\u00eda cardiovascular que hasta ahora resultaban inaccesibles\u201d, explica F. Javier D\u00edez Guerra, autor principal del estudio e investigador del CBM. En concreto, \u201cel nuevo biosensor nos permitir\u00e1 conocer c\u00f3mo aumentos excesivos en la actividad CaMKII contribuyen a la patolog\u00eda de episodios de isquemia en neuronas y c\u00e9lulas card\u00edacas\u201d.<\/p>\n<p>&nbsp;<\/p>\n<h3>Dos prote\u00ednas clave para las neuronas<\/h3>\n<p>&nbsp;<\/p>\n<p>CaMK2rep tambi\u00e9n ha permitido al personal investigador estudiar otra prote\u00edna, llamada Neurogranina, que abunda en las espinas dendr\u00edticas de las neuronas, en especial en regiones clave para la cognici\u00f3n como el hipocampo y la corteza cerebral. Esta prote\u00edna regula c\u00f3mo CaMKII se activa o permanece en reposo. Sus resultados concluyen que Neurogranina limita la actividad de CaMKII y el estr\u00e9s celular favoreciendo la viabilidad y el funcionamiento de las neuronas, esencial para prevenir enfermedades neurol\u00f3gicas.<\/p>\n<p>Las aplicaciones pr\u00e1cticas de la nueva plataforma incluyen una mejor comprensi\u00f3n de los mecanismos de la memoria y el aprendizaje, la posibilidad de avanzar en el estudio de enfermedades neurodegenerativas como el alzh\u00e9imer, o poder analizar c\u00f3mo ciertos f\u00e1rmacos afectan a la actividad cerebral y al sistema cardiovascular.
